What is this plastic piece for?

I've never heard anyone come up with a good answer to that but I've removed them (2 per door) on both our Stangs. IMO, removing them allows water to drain a bit more freely from the door.

As Nuke stated, removing the plastic piece will allow water that gets inside the door to drain more freely.
So why is the plastic piece there in the first place? In theory, it provides a somewhat quieter ride, because you don't have that open slit in the door. Also, it impedes insects that want to crawl inside your door.
If your door only collects a small amount of water, it should be able to drain even with the plastic piece in place. If you get a lot of water inside the door, then the water doesn't drain properly unless you remove the plastic piece.
