No you shouldnt have to. Just pull the plug wire but make sure the engine is cold when doing the work because headers burns arent your friend.

clogged fuel injector possibly. cant always be a spark issue can it? when i had a misfire it was an injector

Heres the firing order for Ford 4.6L, 5.0L HO, 5.4L, 5.8L V-8 engines
