Coolant runs through the egr spacer not the throttle body, and it is to help keep the temps down on the air flowing through the egr spacer. The egr spacer has hot exhaust gasses passing through it , so it needs to be cooled.
The back of the lower intake is a pcv and you need it hooked up other wise you can build up to much crank case pressure and push oil out of the rear main seals as well as other oil seals. Also any oil vapor that escapes from the pcv outlet in the back will eventually coat your nice shiney engine and it will look like hell. The crankcase vent should be on the valve cover/oil filler neck and connect to the throttle body before the throttle plate or it can hook up to the air intake tube someplace.
If it wont run with the pcv line hooked up something is not hooked up right

ok hooked up coolant lines and they are leeking into upper intake, the vacuum is actually sucking the water from the cooling system,so I unhooked the and now have to change oil and repair egr spacer
