Can I run my engine w/ the tranny out?

Can I run my engine w/ the tranny out?
I had a small oil leak around my rear main seal and oil pan gasket, so I took the tranny out and attempted to seal the leak with some silicone (I know that’s not the best solution). I’m not even sure where the oil was coming from exactly, and who knows if or how long the silicone will hold, but would it be ok to run my engine while the transmission is out so I can check for leaks again? Will the motor mounts provide enough support?
Also, I found some oil around my PCV valve and in the vacuum hose that leads to it. How could oil be getting there?
Thanks!

I would bolt the flywheel and bellhousing back up without the clutch. Of course that's added weight on the back of the engine, so maybe I should try to support it with a jack or something? Although there may be too much vibration for that. I guess I'm looking for someone who has tried this without it ending badly before I make some kind of mistake.

Rear main seals are cheap, while your tranny is out and everything why not just install a new one instead of messing with silicone, I can't see that working well at all.
