Have you done a tune up?? Fuel filter, plugs and wires, cap and rotor, you know the basics. Try just regapping the plugs you have now. Something that simple can cause a misfire.

I have a 331 Stroker with Holley upper and lower intakes, Holley heads,E303 cam, 1.6 rockers, 24lb injectors 75mm mass air, 195 feul pump bottom end has been redone as well, 373 gears with a supercharger, MSD coil and distributor butmyMSD is not in right now.Spark plugs have been done over 24's and the gap is .54.Right after I changed my spark plugs I started to get a knock from somewhere up top. It doesn't do it at start up but when I give it a little gas to pull it in the garage. 5spd mind you.Any way it started last week I drove it last week it was fine. Got back in it 20min later andI noticed something wasn't right, it wasmissing and then started pinging soI brought it home and parked it. Thats the story.

Start by checking your timing. Make sure the spout connector didn't fall out or something, its a possibility. But anyway, pull that out when you check your timing, and make sure it is still around 10* or so. Next i would try putting the msd ignition in. You've got a whole lot done to your motor with stock ignition, your supercharger might even be blowing the spark out. Hope that helps
