If you think converting from 3.55 to 4.10 will not effect gas mileage you are on crack!! I don't care how you drive it.
I recall a most that JD made where he posted the gas mileage from his road trip with highway driving using his 4.10s, and his gas mileage wasnt bad at all. Maybe 1 or 2 miles beloew what you got before, but thats nothing big, and if you drice it right your not gonna be gettin that bad of mileage, JD proved that wit his logs of mileage he posted.

I am also thinking about putting 4.10s in my 2000 Mustang GT auto trans...but Is it a significant loss is gas mileage?

To chime in for the sixxers in the crowd-

GTs come with a stock 8.8" rear end where as pre 2010 V6s have a 7.5" rear. IF anyone with a V6 wants to do gears and serious power adders, id highly recommend they buy a complete rear end assembly including a 8.8" rear to handle the extra power.
