Shifting from 1-2-3-D puts no stress on the car. you just tell the trannie when to shift. so unless you keep your car at 5500 RPM's then you will have no problems.
Powerbreaking: this puts stress on the engine and tranny. it wont hurt if you do it occasionally. but if you do it all the time it will wear out your engine and tranny.

The tranny is kind of fly by wire like the throttle... even it you shift it manually the computer still has control... The computer will not allow the shift to take place until the computer thinks it's safe to shift... as demonstrated by the guy in the rental who was complaining about the delay when he was trying to do neutral drops... The computer would shift until the RPMS came down...
