As BeeStang said earlier, the dot is placed on the tire to mark something (it's heaviest point?) for balancing purposes. Accordingly, removal is not recommended.

Once the tire is mounted it's perfectly OK to clean it off. That spot will change as the tire is worn.

You can also ask someone from India what they use to put the black dot on their forehead with and just cover the dot with it. It doesn't seem to wash off on them.
