I've got a '65 with the 200 six cylinder motor. If the car has been sitting for several days, or more, it turns over great, but really needs to crank for a long time before it fires up. If it's been driven that day, it turns over and fires up right away.
I'm thinking I am losing my fuel "prime" and wondering if there's a reasonable fix-new fuel pump, some adjustment, etc. I know I could put in a booster electric pump, but I really don't want to deviate from the stock set-up- this car is 90+ % original
I've also got a bit of hesitation when I go from a stop sign and start up-but gunning the engine a bit seems to help and I suspect that's a separate issue.

You don't say, is this a new issue?
Do you have experience with old cars, or have you only owed and driven newer, fuel injected cars?
What is your start up procedure, when cold and not having been run for days?
