On the passengers side of the t45, there are two plugs. The bottom one is the drain (duh) and the top is the fill hole (also duh). All you need to do is use a ratchet (1/4 drive if i remember correctly) without a socket. Take the bottom plug out, let it drain, put the plug back in, take out the top plug, and fill it till it spills out the hole. It's about 3 1/4 quarts, give or take a little. The only thing you need is a little hand pump or something to get the fluid in. There isnt enough room to just pour straight from the bottle.
If you dont have a pump or dont want to get one, you can remove the shifter and pour it in from the top. But, you'll eff up the gasket and need to put on a new one, so I'd reccomend the pump
And to make this a complete write up, I used Royal Purple Max Automatic Transmission Fluid in my t45. You can also use Royal Purple Synchromax, but the auto fluid is supposedly better, even for use in manuals.

It's a 3/8 ratchet that you need. And this is important: TAKE OUT THE FILL PLUG FIRST!!! You do that because you have to make sure that it comes out. What if you drained it and then the fill plug didn't come out? You'd be screwed.

You can just use RTV to seal up the shifter again. No big deal.

As far as the top plug thing, you could put it in through the shifter....but yeah, thats a good idea if you plan to fill it through the hole.

you can also just use a hose if you have one person to watch for when it fills up under the car. I stuck a hose in the fill hole and ran it up under the hood stuck a funnel in it a filled it up. My buddie just watched for when it started to pour out the fill hole and he screwed in the plug.
