There are so many factors you need to decide on and tell us what you want before we can get anywhere. What kind of mustang is it going in, year and everything. What you want to do with the car; autocross, drag racing, daily driver, etc. What size block; big or small. How long you want the motor to last; a 331 stroker generally lasts longer than a 347 (originally a 302). What end numbers you want for hp and tq. Most importantly what is your budget.

OK first address your first question, best block. There is no for sure answer, but a lot of it can depend on your application. What car is it going in to? 65/66, 67/68, 69/70, or 71/73. All of these have different sized engine bays so a different block can fit better depending.

Also the "best" application could depend on whats available. If you come acrossed on a great deal of an un-bored block then maybe that will be the best just for the money savings.

And as far as what cam and heads, you will get tons of opinions. It all depends on waht you're trying to achieve, and what type of budged you're trying to stay in.

351 Dart block with the 4.125 bore, stock stroke, 375 cubic inches. Scat ultra lightweight forged crankshaft, good lightweight forged pistons(Mahle, CP, Diamond etc) with custom length steel H beam rods from like Manley, ultra light of course, so you get a superlightweight internal, long rod 375W. Brodix Neal BF-301 heads, with the obligatory high end valvetrain, custom ground hydraulic or solid roller camshaft and supporting springs. The Edelbrock intake that works with those heads, good large capacity oil pan with windage tray, and a girdle if you like. High flow water pump, Crane electronic advance controlled dizzy, and digital Hi6 box, underdrive pulleys, long tubes, etc etc. All the goodies obviously, and top it off with a nice Pro Systems carburetor. Be an easy 600hp on pump fuel and have no problem turning 7,000rpm. But you'll end up into it for more than $10,000 just in parts. Then you'd need a drivetrain that can support an engine like that, plus a roll cage if it goes into an early Mustang, since the unibody will want to twist.
