Tuner shift adustment question

I finally got a custom tune for my (mostly stock) 05 V6 auto and finally got to drive it a little this morning (on my way to work). I noticed shiftingis a lotsmoother. However, there's a point somewhere between 40-50 mph when I notice a slight whirring sound. I accelerate just a little and it goes away. But it will happen again when I accelerate to around that 40-50 mph range again. I think it's hitting the 4th gear when it happens. Is it an issue of shift pressure firmness? So if I adjust the firmess (for 4th gear) with the tuner to make it a little more firm, will it likely go away? Thoughts?
